Presynaptic Terminals

چکیده
Fluorescent indicators were used to detect stimulus-evoked changes in presynaptic levels of intracellular sodium (Na1) and calcium (Ca1) in granule cell parallel fibers in brain slices from rat cerebellum. Ca, increased during stimulation, and three exponentials were needed to approximate its return to prestimulus levels. Ca, decayed to -10% of peak levels with T 100 ms, to -1% of peak values with T 6 s, and then returned to prestimulus levels with T 1-2 min. After stimulation, Nai accumulated in two phases; one rapid, the other continuing for several hundred milliseconds. The return of Na, to prestimulus levels was well approximated by a double exponential decay with time constants of 6-17 s and 2-3 min. Manipulations that prevented calcium entry eliminated both the slow component of sodium entry and the rapid component of Nai decay. Reductions of extracellular sodium slowed the rapid phase of Cai decay. These Ca, and Nai transients were well described by a model in which the plasma membrane of presynaptic boutons contained both a sodium/calcium exchanger and a calcium ATPase (Ca-ATPase). According to this model, immediately after stimulation the sodium/calcium exchanger removes calcium from the terminal more rapidly than does the Ca-ATPase. Eventually, the large concomitant sodium influx brings the exchanger into steady-state, leaving only the Ca-ATPase to remove calcium. This perturbs the equilibrium of the sodium/calcium exchanger, which opposes the Ca-ATPase, leading to a slow return of Ca1 and Na1 to resting levels.

Action of brain-derived neurotrophic factor on function and morphology of visual cortical neurons
Brain-derived neurotrophic factor (BDNF) is known to play a role in experience-dependent plasticity of the developing visual cortex. For example, BDNF acutely enhances long-term potentiation and blocks long-term depression in the visual cortex of young rats. Such acute actions of BDNF suggested to be mediated mainly through presynaptic mechanisms. A chronic application of BDNF to the visual cor...

متن کاملRegulation of density of functional presynaptic terminals by local energy supply
Background: The density of functional synapses is an important parameter in determining the efficacy of synaptic transmission. However, how functional presynaptic terminal density is regulated under natural physiological conditions is still poorly understood. Results: We studied the factors controlling the density of presynaptic functional terminals at single dendritic branches of hippocampal n...
متن کاملDevelopmental Up-Regulation of Vesicular Glutamate Transporter-1 Promotes Neocortical Presynaptic Terminal Development
Presynaptic terminal formation is a complex process that requires assembly of proteins responsible for synaptic transmission at sites of axo-dendritic contact. Accumulation of presynaptic proteins at developing terminals is facilitated by glutamate receptor activation. Glutamate is loaded into synaptic vesicles for release via the vesicular glutamate transporters VGLUT1 and VGLUT2. During postn...

Communication between pre- and postsynaptic cells promotes the initial organization of synaptic specializations, but subsequent synaptic stabilization requires transcriptional regulation. Here we show that fibroblast growth factor 22 (FGF22), a target-derived presynaptic organizer in the mouse hippocampus, induces the expression of insulin-like growth factor 2 (IGF2) for the stabilization of pr...
متن کاملLocalized sphingolipid signaling at presynaptic terminals is regulated by calcium influx and promotes recruitment of priming factors.
Activity-dependent changes in presynaptic function represent a critical mechanism by which synaptic strength is controlled. However, how changes in synaptic activity couple to presynaptic components to control synaptic vesicle release and recycling are poorly understood. Sphingosine kinase (SphK) is a sphingolipid metabolic enzyme whose activity-dependent recruitment to membrane regions within ...
